A synthetic peptide conjugated to a carrier molecule is used for immunisation. If you do not have a peptide sequence, you may want to utilise our peptide design service. If you are supplying your own antigen, then simply skip this part and go straight to the antibody production section of the form.NOTE: we offer custom peptide synthesis as a stand-alone service as well.
Peptide/project name
Try keeping the name relatively short or it may be truncated, for instance on labels.Location
The location of the peptide within the protein determines to some extent what the termini should look like and which one should be the conjugation site. If you specify the location we will fill in certain fields below for you.N-terminus
If "Location" of the peptide is specified, you may leave this field as it is. We have only listed the most common N-termini for peptides used in custom peptide polyclonal antisera projects. Many more are available however. Just choose "other" and state what modification you require in the comments box at the bottom of the form.Sequence
In our experience, using peptides of about 15 aa in length will result in excellent antisera. If possible, we would like for the sequence to contain a single cystein located in either one of the termini. This is to facilitate coupling to a carrier molecule and, if your project requires it, a peptide affinity column. Please denote modified amino acids by enclosing them in parentheses; e.g. (pS) for a phosphorylated Serine.C-terminus
If "Location" of the peptide is specified, you may leave this field as it is. We have only listed the most common C-termini for peptides used in custom peptide polyclonal antisera projects. Many more are available however. Just choose "other" and state what modification you require in the comments box at the bottom of the form.Amount
This is the minimum yield of the peptide synthesis. Part of it is used for the peptide-carrier conjugation and, if your project requires it, for the production of a peptide affinity column (2 mg). The remainder of the free peptide is delivered to you.Purity
In our experience of thousands of antiserum projects, we obtain excellent antisera using ImmunoGrade peptides. If your project requires it, you may select higher levels of purity.Peptide-carrier conjugation
There are several coupling strategies available. However, if possible, we do prefer to conjugate via a terminal cystein.Amount
2 mg will be enough for four rabbits using the standard protocol. What material is left when the project is terminated is delivered to you.Carrier

If "Location" of the peptide is specified in the above, or if you don't have a preference regarding conjugation site (N- or C-terminus), you may leave this field as it is.Addition of a terminal cystein
Leave this box checked if we may choose to add a terminal cystein in order to facilitate coupling via its SH-group. In our experience, this strategy of conjugation is preferable. The addition of a terminal cystein for conjugation will not affect the specificity of the antigen.Antibody production
Host
We have only listed the most commonly requested hosts for antibody production. If none of these suit your requirements, just choose "other" and state which host it should be in the comments box at the bottom of the form.Number
Most of our clients choose to use one animal. However, individual animals will respond differently to an immunogen, and the resulting antisera may differ in titre. For this reason one may consider using two or more animals.Cost according to price list
Protocol
Most of our clients choose to follow the standard protocol, making additions and modifications during the course of a project if required. But you are in no way limited to this. If you wish, just choose "custom protocol" and state the details in the comments box at the bottom of the form.Pre-immune sera for screening purposes
If you have reason to suspect cross-reacting issues with the host's pre-immune serum, Innovagen can provide sera from a number of candidates for you to evaluate before any immunisations take place.Cost according to price list
ELISA analysis
The ELISA analysis shows how well the antiserum reacts with the peptide. It is advisable to perform this on the first serum sample to verify that the animal is responding to the antigen. Though the antiserum has to be evaluated in your application, ELISA testing of subsequent samples will allow for easy monitoring of changes in titre.Cost according to price list
Affinity purification
Affinity purification is performed in order to obtain immunogen specific antibodies. The procedure can be performed on any serum sample you like during the course of a project. Included are IgG purification, the production of a peptide affinity column, and purification using this affinity column. Both IgG purification and peptide affinity column production are also available as stand-alone services.Cost according to price list
IgG purification
IgG purification (or its equivalent depending on species of the host) is performed in order to reduce background signal caused by other proteins and molecules present in the crude sample. Tick this box if you would like Innovagen to IgG purify your antiserum samples.Cost according to price list
Aliquotation
We recommend that antiserum is aliquoted. Working with aliquots limits the extent of the damage in the event of an accidental contamination. It also helps keeping repeated thawing and freezing to a minimum. Save time and effort: just tick this box and Innovagen will aliquot your antiserum for you.Cost according to price list
Sterile filtration
Sterile filtration is performed in order to remove any microbial contamination that may be present in the sample. Tick this box if you would like Innovagen to sterile filtrate your antiserum samples.Cost according to price list
Azide addition
The addition of sodium azide prevents microbial growth upon accidental contamination of your antiserum. Tick this box if you would like Innovagen to add sodium azide to your antiserum samples.Cost according to price list
